More collagen: shown in dishes, not faces
Most tightening claims say a peptide makes skin produce more collagen. For KTTKS, the core of Matrixyl, that has been shown in cultured cells [10], and for GHK-Cu in cultured fibroblasts [11]; the topical trials we cite measured wrinkles and fine lines, and their abstracts report no collagen measurement [1][6].
- In vitro (cells) The collagen effect behind Matrixyl comes from cell studies: its core peptide without the fatty tail, KTTKS, raised collagen production in cultured fibroblasts and other connective-tissue cells. The two Matrixyl trials we cite measured wrinkles (image analysis, expert grading, photos) and, in the smaller one, skin hydration and elasticity; neither abstract reports a collagen measurement.[10][1][6]
- Animal GHK-Cu raised collagen in cultured fibroblasts and in rat wound chambers. A 2025 review notes a "surprising absence of clinical studies" of topical GHK-Cu.[11][12][13]
- In vitro (cells) The mechanism cited for palmitoyl tripeptide-5 (SYN-COLL, as sellers describe it) comes from lab work on how thrombospondin-1 sequences activate TGF-beta; that paper does not test palmitoyl tripeptide-5 itself. The one human study we found used it in nanoparticles with lactoferrin and recombinant collagen, and its abstract states no control group.[14][2]

Oral collagen and elasticity
A 2025 meta-analysis pooled 23 randomized trials of collagen supplements on skin aging [9]. The trouble is who paid for them.
Sagging after weight loss
Fast weight loss can leave facial skin looser, the pattern called "Ozempic face". This is about lost facial volume and fat, and we found no topical peptide tested for it.

Does Matrixyl work?
In one 12-week trial, 93 women had significantly fewer fine lines than with the same moisturizer without it. A smaller 8-week trial of 21 women also favored it.[1][6]
How long does Matrixyl take to work?
Its trials ran 8 and 12 weeks. We found no trial of shorter use.[1][6]
What does palmitoyl tripeptide 5 do?
The mechanism cited for it comes from lab work on thrombospondin-1 sequences that did not test palmitoyl tripeptide-5 itself, and a 2021 review found that support for it and six other cosmetic peptides sits mostly in patents and supplier brochures rather than randomized placebo-controlled studies.[19][14]
Does argireline cause sagging?
We found no study showing that. In lab tests argireline barely got past the skin surface, and a 2025 review says whether it reaches or relaxes facial muscles is uncertain.[20][21]
How long does it take for collagen to work on skin?
The trials cited here point to about 8 to 12 weeks: an 8-week trial in 69 women, and a meta-analysis that reports 90 days of intake. In a 2025 meta-analysis, the effect was absent in trials without pharmaceutical-company funding and in high-quality trials.[18][7][9]
